زمینه و هدف: کلرپیریفوس یکی از پر مصرف ترین سموم ارگانوفسفره است که به‌طور عمومی در دنیا استفاده می‌شود. این سم علاوه بر مهار آنزیم کولین استراز، در القاء استرس اکسیداتیو و افزایش اکسیدآن‌ها نیز نقش دارد. از طرفی عصاره گیاه رزماری یک ماده آنتی‌اکسیدانی است و نقش محافظت کننده ای در برابر رادیکال های آزاد ایفاء می کند. بنابراین هدف از انجام این مطالعه، بررسی پیشگیری از آسیب کبدی ناشی از مواجهه با سم تجاری کلرپیریفوس در نتیجه مصرف عصاره رزماری به دلیل خاصیت آنتی‌اکسیدانی این گیاه می‌باشد.
روش بررسی: در این مطالعه از 30 راس موش صحرایی نر نژاد ویستار با محدوده وزنی 200-220 گرم استفاده شد. در ابتدا حیوانات به‌طور تصادفی به پنج گروه (هر گروه 6 سر) تقسیم شدند. پنج گروه شش تایی شامل، گروه کنترل تیمار با آب مقطر به صورت تزریق درون صفاقی، گروه مواجهه با کلرپیریفوس به صورت تزریق درون صفاقی (دوز mg/kg 5/13)، گروه مواجهه با عصاره رزماری به صورت خوراکی و توسط گاواژ (غلظت mg/kg100)، گروه مواجهه توام با سم به صورت تزریق درون صفاقی (دوز mg/kg 5/13)و رزماری به صورت خوراکی و توسط گاواژ (غلظت mg/kg 100) ، گروه مواجهه همزمان سم به صورت تزریق درون صفاقی و رزماری به صورت خوراکی و توسط گاواژ (غلظت mg/kg 200 خوراکی)، به‌عنوان گروه‌های مورد مطالعه انتخاب شدند. بعد از 30 روز متوالی، موش‌ها وزن و کشته شدند و بافت کبد جهت بررسی‌های هیستوپاتولوژی و تغییرات سطح مالون دی آلدئید و فعالیت آنزیم گلوتاتیون پراکسیداز مورد بررسی قرار گرفت. جهت سنجش سطح مالون دی آلدئید، دستگاه الایزا ریدر مدل بیوتیک بر روی طول موج 535 نانومتر و جهت سنجش سطح فعالیت آنزیم گلوتاتیون پراکسیداز، دستگاه بر روی طول موج 412 نانومتر تنظیم شد. سپس داده‌های حاصل توسط نرم‌افزار آماری SPSS 22 و روش آزمون آنالیز واریانس یک‌طرفه One-way ANOVA و به دنبال آزمون آماری Tukey مورد تجزیه‌وتحلیل آماری قرار گرفتند و تفاوت میانگین‌ها در سطح P<0.05 معنی‌دار در نظر گرفته شد.
یافته ها: طبق تست های آماری گروه مواجهه یافته با عصاره رزماری نتایج مشابه گروه کنترل یافت شد اما در گروه مواجهه یافته با کلرپیریفوس این سم به‌طور معنی‌داری باعث تغییر در میزان فعالیت آنزیم گلوتاتیون پراکسیداز و مالون دی آلدئید نسبت به گروه کنترل و همچنین آسیب‌هایی از جمله آشفتگی در چیدمان سلول‌ها، افزایش فضای سینوزوئیدها، نکروز سلول‌های کبدی و ادم سیاهرگ مرکزی در بافت کبد می‌شود در حالیکه مواجهه همزمان با کلرپیریفوس و عصاره رزماری به‌طور معنی‌داری باعث بهبود فعالیت آنزیم گلوتاتیون پراکسیداز و سطح مالون دی آلدئید و  همچنین کاهش آسیب ناشی از کلرپیریفوس می‌گردد.
نتیجه گیری: با توجه به نتایج فوق به نظر می رسد که سم کلرپیریفوس به واسطه تولید رادیکال‌های آزاد، تنش اکسیداتیو و تضعیف دفاع آنتی‌اکسیدانی باعث آسیب سلولی و تغییرات ساختاری در بافت کبد موش‌های صحرایی می‌شود. درحالی‌که عصاره رزماری به دلیل دارا بودن ترکیبات آنتی‌اکسیدان باعث افزایش فعالیت آنتی‌اکسیدانی، کاهش میزان پراکسیداسیون لیپیدی و محافظت در برابر آسیب کبدی ناشی از ماده سمی کلرپیریفوس می‌گردد.
